温度是影响VIGS(病毒诱导的基因沉默, virus induced gene silencing)对目标基因沉默效率的重要因素之一。【目的】为了明晰VIGS-TRV体系临界高温对NtPPO基因沉默的效果,【方法】本研究利用NtPPO7和NtPPO8的VIGS载体,探究在25℃、40℃、43℃、46℃下,NtPPO7和NtPPO8基因表达量以及TRV(烟草脆裂病毒,Tobacco rattle virus)浓度的变化。【结果】40℃时的VIGS基因沉默效率低于25℃时的沉默效率,43℃和46℃时沉默效果消失;40℃温度处理烟株后使烟苗置于25℃下生长,随着时间的推移沉默效果仍低于25℃处理的烟株。25℃处理的烟株随着时间增加基因沉默效果逐渐降低。【结论】VIGS-TRV体系临界高温为40℃~43℃,高于43℃,目标基因沉默效果不可逆消失。研究为VIGS-TRV体系大田生产应用及后期明确临界温度提供了借鉴意义。
