This work presents a demonstrative and educational study illustrating intracellular reactive oxygen species (ROS) generation using the 2′,7′-dichlorodihydrofluorescein diacetate (DCFH-DA) fluorescence assay. The document describes the biological basis of ROS production, the principle of the DCFH-DA assay, and its application in detecting oxidative stress following hydrogen peroxide exposure in an in-vitro cell culture model. The study is confirmatory in nature and is intended to reinforce established concepts in redox biology rather than to report novel mechanisms or validated protocols. It serves as an instructional example for understanding experimental design, data interpretation, and commonly used laboratory techniques in undergraduate medical education. This work is provided for academic and educational purposes only. It does not propose a standardized, optimized, or clinically validated assay and is not intended for diagnostic, therapeutic, or clinical decision-making use.
