Plasmalemma vesicle associated protein (PLVAP) is involved in the formation of diaphragm in cellular structures like fenestrae, caveolae, and transendothelial channels. Scientific reports have primarily focused on PLVAP and its association with Caveolin-1 in fixed cells. Here, we have explored the dynamics of PLVAP in live endothelial cells at the plasma membrane with the help of fluorescence and interference reflectance microscopy. In addition to PLVAP, we have also focused on the relation of PLVAP with Clathrin, Caveolin1, filamentous actin, and exosomal marker CD63. Interestingly, we observed a significant increase of PLVAP at the plasma membrane during live cell imaging. Also, generally PLVAP is known to associate with Caveolin1, but our data showed huge clusters of PLVAP that did not colocalize with Caveolin1. Based on our preliminary live imaging analysis, we infer that huge amount of PLVAP is targeted to the plasma membrane, and we speculate that it might be involved in other cellular functions than diaphragm formation.
