Abstract Global paediatric immunization coverage for the pentavalent vaccine (Diphtheria , Tetanus, Pertussis, Hepatitis -B, Haemophilus influenzaetype-b type-b) has compounded by the physiological “injection burden” and magnified by the logistical constraints of parental administration. This perspective explores the transformative potential of a liquid oral syrup stabilized by a pH – responsive polymeric matrix. By involving an acid resistant shield of Eudragit L 100 with mucoadhesive chitosan nanoparticles, we propose a mechanistic framework to preserve antigenic integrity against gastric proteolysis while ensuring targeted delivery to the intestinal Paeyer’s patches. We argue that transitioning to a non- invasive, oral – route delivery system is a critical evolution required to achieve Universal health equity and overcome vaccine hesitancy in the health care system(CDC,2025 )
