Wireframe RNA nanostructures with defined geometry are desirable, but bendable RNA edges can lead to structural deformation. Here, we present our design of single-stranded RNA Origami with bundled duplexes as edges. With dual duplex bundles as the wireframe edges, a specific geometry results for our RNA constructs. We construct a number of complex RNA polygons and grids with high yield and quality. Our approach can further expand the RNA design possibilities and enable downstream biomedical applications.
