Xanthoceras sorbifolia Bunge is a traditional medicinal plant with multiple application values. Yi Niao Ting Capsule, which is made from the seed meal of Xanthoceras sorbifolia Bunge, has the effect of tonifying qi and spleen, tonifying kidney and reducing urine. In this paper, the chemical composition and pharmacological mechanism of Xanthoceras sorbifolia Bunge seed meal were studied by modern scientific and technological means combined with traditional Chinese medicine theory. to investigate the effect of Xanthoceras sorbifolia Bunge seed meal extract on polyuria in rats with kidney deficiency syndrome, and compare the chemical constituents and pharmacodynamics of different extracts. The polyuria model of kidney qi deficiency rats was constructed with kanamycin sulfate. After drug intervention, the changes in body temperature, body weight and activity of rats were observed, and 24 h urine was collected. The contents of adrenocorticotropic hormone (ACTH), cortisol (CORT), cyclic adenosine monophosphate (cAMP), protein kinase A (PKA) in serum and aquaporin 2 (AQP2) in kidney tissue were detected by ELISA. The kidney mass was weighed to calculate the organ index and the renal tissue was stained with HE to observe its changes. Ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS) was used to analyze chemical components. compared with the blank group, the body weight of the rats in the model group was significantly decreased ( P < 0.05), and the urine volume was significantly increased ( P < 0.05). The contents of ACTH, CORT, cAMP, PKA in serum, and AQP2 in kidney tissue were down-regulated to varying degrees ( P < 0.05). The kidney index was significantly increased ( P < 0.05), and the kidney tissue structure was severely abnormal. After drug intervention, the seed meal extract group could significantly increase the body weight of rats ( P < 0.01), reduce urine volume ( P < 0.01), up-regulate the contents of ACTH, CORT, cAMP, PKA, and AQP2 ( P < 0.05), and improve the degree of renal tissue damage. A total of 106 compounds were identified from the seed meal extract, and flavonoids, phenols, and fatty acids were the main components. Xanthoceras sorbifolia seed meal extract can regulate the body's water metabolism by activating cAMP-PKA-AQP2 pathway activity and HPA axis system, and play a role in tonifying the kidney and reducing urine. The polyphenols in the seed meal of Xanthoceras sorbifolia may be related to the efficacy.
