Export A new classification of medulloblastoma was introduced in 2016 and beyond. This classification was designed to accommodate the incorporation of updated data on the biology, genomics, and clinical outcome of medulloblastoma. As a result, medulloblastomas were classified according to molecular features (also known as “genetically defined” categories) as well as histological features (also known as “histologically defined” categories). Recent groups that have been “genetically defined” include those that are WNT-activated, SHH-activated TP53 wildtype, SHH-activated TP53-mutant, and non-WNT/non-SHH. The primary objective of this article was to provide a comprehensive assessment of the most current update that was included in the World Health Organization’s classification of medulloblastomas in 2021. This update included the layered or integrated diagnostic approach, which incorporates both molecular and histological categories within a single diagnostic report.
