Abstract Background: Asthma is a chronic inflammatory respiratory disease associated with changes in airway structure and inflammation. Eucalyptol has demonstrated immune modulatory activity as well as anti-inflammatory and relaxant properties on airway smooth muscle. Quercetin is thought to cause relaxation of airway smooth muscle. Objectives: The purpose of this study was to examine the potential combined anti-asthmatic effect of eucalyptol (1,8-cineole) and quercetin in Swiss Albino mice. Methods: The study investigated the effects of quercetin (10 mg/kg) and eucalyptol (30 mg/kg), using two asthma models; ovalbumin-induced asthma and milk-induced asthma. The potential therapeutic effects of quercetin and eucalyptol when administered as treatments for both asthma models were evaluated. The ovalbumin model was based upon sensitization of the animals with ovalbumin, whereas the milk model was based on sensitization of the animals with milk. Parameters such as airway inflammation, lung function, and inflammatory markers were evaluated. For in-vivo research, Swiss albino mice were split up into 4 groups of 6 animals each. Findings: The findings demonstrated that the combined treatment decreased the animal’s overall leukocyte and eosinophil numbers. Facilitated changes in haematological parameters, and decreased Th2 cytokine production (IL 4, TNF-α, IL-13) levels that is induced by Ovalbumin (OVA) in lungs and the serum level of total IgE. The combined action of Eucalyptol and quercetin decreased inflammatory responses brought on by OVA influx and ultrastructural abnormalities, according to histopathological evaluation. Application: The findings of this investigation demonstrate that Eucalyptol and Quercetin together possess anti-asthmatic effects through suppressing Th2 triggered immune response and possessing an both an anti-allergic and anti-inflammatory action.
