Abstract Blastmycetin G (1), a 5-hydroxy derivative of teleocidin B’s, was isolated from Streptomyces blastmyceticus NA34-17. The 5-hydroxy group fixes exclusively the nine-membered lactam ring into a sofa (trans-amide) conformation. The significantly weaker binding affinity for PKCα and PKCδ-C1 domains (400–800 times less than teleocidin B-4) supports that the sofa conformer is biologically inactive.
