Objective Leukocyte‐ and platelet‐rich fibrin (L‐PRF) has emerged as a bioactive scaffold with regenerative and immunomodulatory properties, though its antimicrobial potential remains underexplored. This study aimed to evaluate in vitro the inhibitory effect of L‐PRF membranes on Streptococcus mutans and Candida albicans in monoculture and coculture conditions. Materials and Methods L‐PRF membranes were prepared from venous blood of healthy donors via centrifugation at 2400 revolutions per minute (rpm) for 12 min and segmented into head, body, and tail regions. Reference strains S. mutans American Type Culture Collection (ATCC) 25175 and C. albicans ATCC 10231 were cultured on Müller–Hinton agar, and antimicrobial activity was assessed using agar diffusion assays under microaerophilic conditions (37°C, 5% CO 2 ). Inhibition zones were measured post‐24 h incubation. Results L‐PRF membranes showed no inhibition against S. mutans in monoculture but exhibited significant activity against C. albicans (head and body segments). In coculture, both segments inhibited microbial growth. Inhibition halos were larger for C. albicans than S. mutans , with complete growth suppression observed beneath the membranes. Conclusions L‐PRF demonstrates selective antimicrobial effects, particularly against C. albicans , suggesting its potential as an adjunct in oral infections. Further studies should clarify the mechanistic pathways and optimize protocols for clinical translation.
