To identify potential target genes that YY1 mediate copper ionophore-induced cell death in glioblastoma(GBM) LN229 cells. The knockdown of YY1 downregulated glycolytic process and copper ion transmembrane transporter activity by The Gene Set Enrichment Analysis (GSEA) . The candidate YY1 target genes including the cuproptosis-related genes (CRG), glycolysis and redox-metabolism-regulating genes are identified, such as ALDH1A3, SLC31A1, and metallothioneins (MTs: MT2A and MT1E), which are key downstream target genes of YY1 closely correlated with copper ionophore-induced cell death. Our data showed YY1 prevents copper ionophore-induced cuproptosis via regulating expression of ALDH1A3 and MTs genes.
