This review critically examines the evidence for beta-glucan-containing foods and supplements – brewer’s yeast, oats, and barley – as galactagogues for breastfeeding women. We demonstrate that beta-glucans from these three sources are structurally distinct (cereal vs. yeast), interact with different biological receptors, and that the prolactin-stimulating mechanism attributed to beta-glucan has only been demonstrated in vitro and via intravenous injection but never through oral consumption. The first human randomized controlled trial of brewer’s yeast (Jia et al. 2025) showed no objective effect on milk supply. We further identify an unexamined safety dimension: brewer’s yeast acts as a histamine liberator and contains biogenic amines (tyramine, putrescine) that compete with histamine for diamine oxidase (DAO) clearance, potentially worsening symptoms in postpartum women with histamine sensitivity. Computational modeling of the human DAO active site and experimental validation studies are underway. This work proposes a multi-dimensional framework for evaluating galactagogues beyond single-axis efficacy assessment.
