Abstract: BCR-ABL tyrosine kinase is a well-recognized target for antileukemic therapy. Four generations of BCR-ABL tyrosine kinase inhibitors have been employed to treat leukemia; however, their efficacy is often limited due to resistance exhibited by leukemic cells. This resistance arises from mutations and overexpression of the BCR-ABL gene, highlighting the ongoing need for novel BCR-ABL kinase inhibitors. Plant extracts and phytoconstituents have demonstrated both BCR-ABL kinase inhibitory activity and potent antileukemic effects. Several studies have also investigated the specific inhibitory activities of phytoconstituents against BCR-ABL and the downstream pathways they modulate. This review summarizes in vitro and preclinical studies on various classes of natural products and semisynthetic compounds that inhibit BCR-ABL and its associated signaling pathways.
