ABSTRACT Multiple organ injury including the intestines, liver, and central nervous system is a prevalent consequence of chronic alcoholic intoxication (CAI). Compelling evidence from recent studies underscores a central role for the intestines and their microbiota in mediating interactions between the gut and the liver or the brain. In this article, the effects of polysaccharides from Eucommia ulmoides Oliv. leaves (EULP) on chronic alcoholic tissue injury along the gut–liver or gut–brain axis were studied. Results demonstrated that EULP alleviated gastric mucosal damage, endotoxemia, epithelial tight junction disruption (Occludin and Claudin‐1), and mucosal barrier dysfunction in CAI mice. Gut microecology analyses indicated that EULP modulates CAI‐induced changes in the diversities and abundances of gut microbiota. Furthermore, EULP attenuated chronic alcoholic liver inflammation and oxidative stress, concomitantly improving neurotransmitter secretion while reducing neuroinflammation and oxidative stress in brain tissue. These findings suggest that EULP's ability to alleviate alcohol‐induced intestinal epithelial barrier dysfunction and modulate the intestinal microbiota plays a crucial role in the mechanism underlying its amelioration of CAI induced tissue injury at the gut–liver or gut–brain axis.
