Here, we present an in vitro assay for quantifying fibroblast-mediated contractile dynamics using a three-dimensional collagen plug. We describe steps for suspending fibroblasts in neutralized collagen type I, polymerizing to form uniform collagen plugs, and stimulating contraction by growth factors to induce myofibroblast activation. We detail procedures for measuring contraction of the collagen plug by quantifying its area and assessing matrix remodeling. This protocol allows for the screening and assessment of potential therapeutic targets for preventing fibrosis.
