Background: Gastritis is a common health condition that may progress to peptic ulcers, causing bleeding, perforation, peritonitis, or death. Cytokines, including interleukin-8 (IL-8) and interleukin-10 (IL-10), are key mediators in the inflammatory process. Hyperbaric oxygen therapy (HBOT) is suggested to enhance mucosal healing. This study aims to evaluate IL-8 and IL-10 expressions in aspirin-induced gastric mucosal inflammation in Wistar rats after combination therapy of HBOT and omeprazole. Methods: An experimental study with posttest-only control group design was conducted in 32 male Wistar rats, divided into 4 groups: negative control (group 1), positive control (group 2), group 3 (aspirin induction at 30 mg/kgBW/day, followed by omeprazole 0.72 mg/kgBW/day for 10 days), and group 4 (aspirin induction at 30 mg/kgBW/day, followed by the combination of omeprazole 0.72 mg/kgBW/day and HBOT 2.4 ATA). The expression of IL-8, IL-10, and histopathological inflammation in each group were evaluated and analyzed. Results: Combination of HBOT with omeprazole for 10 days reduced inflammation (p < 0.001) and reduced IL-8 expression (p < 0.001), significantly. This combination therapy (group 4) also improved IL-10 expression (p = 0.004). Group 3 and group 4 showed significant differences in decreased inflammation (p = 0.012) and decreased IL-8 expression (p = 0.001). However, no significant difference was observed between group 3 (omeprazole therapy only) and group 4 (combination therapy), in the increase in IL-10 expression (p = 0.051). Conclusion: HBOT in combination with omeprazole enhances gastric mucosal healing in aspirin-induced injury by reducing inflammation and IL-8 expression while promoting IL-10 activity. This suggests that HBOT may serve as a beneficial adjunct therapy for gastritis.
