Abstract Background: The field of drug delivery has witnessed remarkable advancements aimed at optimizing therapeutic outcomes and reducing adverse effects. In this study, we examined the ameliorative effect of ultrasound waves in combination with Functionalized Carbon nanotubes -encapsulated Dacarbazine (fSWCNT-DTIC) in a murine B16F10 melanoma model. Materials and Methods: The Preparation of single-walled carbon nanotube encapsulated Dacarbazine (fSWCNT-DTIC) accrued at several steps. The melanoma tumor model was induced by injection of B16F10 cells subcutaneously into the right shoulder of c57bl/6 mice. After the tumor reached a volume of 100mm 3 each group was treated locally with 60 μL of either vehicle (normal saline), Dacarbazine, Carbon nanotube, Ultrasound, or a combination of them. Tumor Growth inhibition, systemic toxicity, and also liver and lung histopathological changes were assayed in different groups. In all analyses, a significance level of 0.05 was deemed appropriate. Result: The highest Tumor Growth inhibition (%88.21) was seen in the group that received single-walled carbon nanotubes encapsulated Dacarbazine and also exposed to ultrasound radiation. In various groups, there was no noticeable variance in weight loss at the experimental period. Combination therapy improved the chaotic condition, inhibited the spread of cancer cells in the lungs, reduced severe inflammation in the lung lobes and reversed abnormal patterns in the liver. Conclusion: The combination of ultrasound and SWCNT-encapsulated Dacarbazine, could make a significant difference in the treatment process and improve the treatment outcomes for cancer patients.
