Scleraxis is a basic helix-loop-helix transcription factor, which is a key regulator of musculoskeletal connective tissue development. It is essential for the differentiation and maturation of tendons and ligaments throughout embryogenesis. In their work, Chisa Shukunami, Shinsei Yambe, Kenta Sumiyama and colleagues identify a 343 bp conserved enhancer that contributes to the spatiotemporal regulation of Scleraxis expression and deltoid tuberosity morphogenesis during limb development.
