Key Points

  • On-treatment analysis indicates changes in CD8+ T-cell populations responding to treatment.
  • Multiplex immunofluorescence was used to assess T-cell dynamics in bladder tissue samples from two responders.
  • Phase IB NABUCCO study provides insight into the effectiveness of ipilimumab combined with nivolumab.
  • Single-cell RNA-sequencing further characterizes the therapeutic response and dynamics of specific T-cell subtypes.

Abstract

Treatment dynamics of CD8+TCF7+ T-cells in two responders to ipilimumab-high. a. Baseline bladder tissue stained with a multiplex immunofluorescence T-cell panel (DAPI (blue), PanCK (white), CD8 (red), TCF7 (yellow)) for responder #1 (ypT0N0) to high-dose ipilimumab plus nivolumab. b. On-treatment resected bladder tissue stained with a multiplex immunofluorescence T-cell panel for responder #1 (ypT0N0) to high-dose ipilimumab plus nivolumab; c. Baseline bladder tissue stained with a multiplex immunofluorescence T-cell panel for responder #14 (ypT0N0) to high-dose ipilimumab plus nivolumab; d. On-treatment resected bladder tissue stained with a multiplex immunofluorescence T-cell panel for responder #14 (ypT0N0) to high-dose ipilimumab plus nivolumab. The on-treatment tissue of both patients was used for single-cell RNA-sequencing to further characterize the CD8+TCF7+ cells.
