Key Points

  • To develop a tool that optimizes mRNA sequences through synonymous codon exchange for improved protein production.
  • Developed OCTOPOS.jl as a Julia package for codon optimization.
  • Implemented a web interface for user accessibility.
  • Focused on optimizing translation dynamics without altering protein structure.
  • Increased protein yield by optimizing codon usage in different host organisms.
  • Demonstrated effective alteration of translation dynamics through codon exchange.

Abstract

The genetic code of organisms connects which codons will be translated to which aminoacid. There are multiple codons which encode the same aminoacid, but they have differing translation dynamics. This can be used to alter the translation dynamic of a mRNA without changing the resulting protein. OCTOPOS.jl is a Julia package for optimizing mRNA sequences by synonomous exchange of codons. This can be used for example to increase protein yield in different host organisms.A web interface is available at https://octopos.cell.uni-hannover.de.
