Abstract

目的 比较参环毛蚓药用部位与非药用部位提取蛋白的溶栓活性与功能,为广地龙中药废弃物资源再利用提供科学依据。 方法 通过对参环毛蚓药用与非药用部位提取的蛋白进行BCA法蛋白含量测定、SDS-PAGE法蛋白分子量分布测定、琼脂糖-纤维蛋白原平板法测定蛋白纤溶活性和SOD抗氧化酶活性的测定,分析判断两组提取蛋白的功能及溶栓活性。 结果 药用部位与非药用部位除提取蛋白的提取率与蛋白含量无显著性差异,纤溶活性和抗氧化能力非药用部位皆显著性高于药用部位,且对照蚓激酶分子量分布,结合SDS-PAGE结果与纤溶活性测定表明具有纤溶活性蛋白的分子量主要集中在25 ~33 kDa,此区间蛋白相对占比均大于50%。 结论 药用地龙参环毛蚓的非药用部位作为中药废弃物,以溶栓作用为主的纤溶活性及其血栓形成成负相关的SOD酶活性都远大于药用部位,是优于药用部位提取纤溶活性蛋白的组织部分。该研究为其中药废弃物资源的利用提供理论依据,为相关活性蛋白的可提取资源进行补充,助力参环毛蚓产业的可持续发展。
