This study establishes a rapid diagnostic platform using UPLC-MS/MS to quantify urinary tract infection biomarkers within 8 minutes. Targeting phenylacetylglycine (PAG, E. coli) , cyclic diguanylate monophosphate (c-di-GMP, K. pneumoniae ), and pyocyanin (PYO, P. aeruginosa ), we optimized a 3-min sample preparation and 5-min chromatographic separation. The portable Waters Xevo TQ-S micro system achieved detection limits of 0.008–0.03 μmol/L and precision 93% concordance with culture and 42% higher mixed-infection detection (71 vs 50), enabling pathogen-directed therapy within 45 min of admission.The rapid diagnosis facilitated precision nursing interventions including targeted catheter care and infection control measures.
