In this work, a benzo-imidazole-derived fluorescent probe BIENBD-GSH was developed for monitoring the hypoxic alleviation with dexmedetomidine protection via the glutathione (GSH) detection. The structural modification of moving the recognition site and extending the meta-group was performed. The normoxia and hypoxia conditions did not affect the absorption and fluorescence variation during the response of BIENBD-GSH to GSH. BIENBD-GSH exhibited a dose-dependent response to the GSH level to report the fluorescence signal at 500 nm under the excitation of 365 nm. The structural modification introduced the advantages of higher synthetic yield and emission red-shift, with the retention of a wide linear interval and relatively high sensitivity. Other advantages of BIENBD-GSH included stable performance in various working conditions, high selectivity towards GSH, and low cytotoxicity upon living pulmonary cells. BIENBD-GSH achieved monitoring the intracellular GSH level, while for the hypoxia-related situations, both the post hoc recovery and prior dexmedetomidine protection were visualized. This work provided some useful information on the pathological processes in clinical operations and the nursing schedules.
