The possibility of determining water-soluble formazans of tetrazolium dyes WST-8 and XTT by Raman spectroscopy was studied. It was shown that XTT does not have a pronounced Raman spectrum and is therefore not suitable for determination by this method. Raman spectroscopy was used to compare WST-8 formazan and water-insoluble MTT formazan. It was demonstrated that WST-8 can be determined with greater sensitivity. However, the signal intensity in the presence of Escherichia coli cells was higher for MTT formazan, with negligible differences in the detection limit of cells between the two reagents.
