Abstract

In this comprehensive study, we delve into the complex interplay between the intratumoral microbiome and tumor dynamics in melanoma, with a special focus on the role of Pseudomonas aeruginosa extracellular vesicles (EVs). Employing advanced spatial transcriptomics alongside traditional microbiome analysis, we characterize the diverse microbial landscape within melanoma tumors and examine the interactions between these microbes and host tumor cells. A significant finding of our research is the specific impact of Pseudomonas aeruginosa EVs on melanoma cell behavior. Our experiments reveal that while these EVs do not significantly alter the proliferation of A375 melanoma cell lines under the tested conditions, they notably enhance cell migration. This is evidenced by Transwell migration assays and wound healing assays, where EV-treated groups demonstrated increased migratory capacity compared to controls. Such findings suggest a pivotal role of these bacterial vesicles in modulating the tumor microenvironment, particularly by promoting cell movement and potentially facilitating tumor invasion and metastasis. Moreover, our study highlights the potential of Pseudomonas aeruginosa EVs to modulate inflammatory responses within the tumor microenvironment, thereby influencing tumor-immune interactions. These vesicles act as carriers of biologically active molecules, mediating complex interactions between intratumoral bacteria and host cells, and underscoring the intricate relationship between the intratumoral microbiome and tumor dynamics. In conclusion, this research sheds light on the multifaceted influence of the intratumoral microbiome, especially Pseudomonas aeruginosa EVs, on melanoma biology. By providing new insights into microbial-host interactions within the tumor microenvironment, our study opens avenues for exploring innovative therapeutic strategies targeting these microbial components in melanoma treatment.
