, underscoring the importance of balanced polyglutamylation for morphogenesis and cell cycle progression in kinetoplastids. IMPORTANCE: is used as a model system to examine the functional roles of two microtubule polyglutamylases, TTLL4C and TTLL6B. The findings reveal that these enzymes are essential for maintaining cytoskeletal integrity, cell morphology, organelle positioning, and normal cell growth. Collectively, this work advances our understanding of microtubule regulation and highlights the broader cellular functions governed by tubulin polyglutamylation.
