Abstract

We present the first geometric pharmacology atlas unifying Traditional Chinese Medicine (TCM) Four-Nature compounds and Western drug classes on a single QLTE x GII coordinate system. Forty-two compounds (15 TCM, 27 Western spanning six pharmacological classes: antipyretic/analgesic, opioid, anesthetic, antidiabetic, antihypertensive, muscle relaxant) were analyzed using Lo-Shu Transition Energy (QLTE), Geometric Instability Index (GII), TPSA, and logP. Drug classes segregate geometrically consistent with clinical pulse effects, with TPSA serving as the primary Yang/Yin discriminator. Three pharmacological anomalies (Ketamine in Yang zone, Metformin with highest GII, Cyclobenzaprine with TPSA=3. 2) provide mechanistic validation. A five-dimension pulse effect matrix (Rate/Force/Depth/Smoothness/Width) provides a clinical reference for integrative practitioners. The framework clarifies that geometric position encodes membrane penetration capacity while pulse effect encodes target-mediated hemodynamic change, resolving apparent paradoxes such as NSAIDs appearing in the Yang geometric zone while clinically reducing Yang-excess pulse patterns.
