Türkiye is an important genetic center for rose species, with approximately 45 different rose varieties found throughout the country. The green rose, native to China and scientifically known as Rosa chinensis viridiflora, grows in the Halfeti district of Şanlıurfa and is known as a rare ornamental plant synonymous with the region. Due to its limited distribution, the conservation and sustainable propagation of this variety are of great importance. Optimization of culture media is critical in the successful in vitro propagation of ornamental plants. In this study, different rooting media (R1: 0.5 mg L⁻¹ IBA + 25 µM SNP, R2: 0.5 mg L⁻¹ NAA + 25 µM SNP, and R3: 0.5 mg L⁻¹ IBA + 0.5 mg L⁻¹ NAA + 25 µM SNP) were used to determine the effects of different culture media (T1: Murashige and Skoog (MS), T2: Driver and Kuniyuki Walnut Medium (DKW), T3: Woody Plant Medium (WPM), T4: Nitsch and Nitsch (NN) on shoot development and rooting success of green rose explants. The study evaluated parameters such as the percentage of explants forming shoots, shoot length, leaf number, and node number, as well as root diameter, root length, root number, and rooting rate. T4 medium showed the highest explant formation rate (22%), while T1 medium was superior in terms of shoot length (1.67 cm), number of leaves (11.27 per plantlet), and number of nodes (4.84 per plantlet). On the other hand, R1 medium provided the highest values in terms of root length (4.75 cm) and number of roots (2.75 per plantlet), while R3 medium increased root diameter (0.79 mm). Circular heatmap based on hierarchical clustering analysis comprehensively revealed the relative effects of the treatments. These findings contribute to the development of in vitro propagation protocols for green roses.
