Abstract

This technical report defines the U131-Kampo Generalized Structural Screening Framework, extending the earlier active6 Kampo structural classifier into a cross-corpus botanical and compound-network governance model. The framework preserves active6 + official148 as the committed Kampo baseline while maintaining official180/official200 as candidates, shadow294 as a non-committed formula ledger, Herb500 as a governed material ledger, and Herb1000 as a stress-only set.The report formalizes normalized material records, compound and biosynthesis proxies, target-protein proxies, a non-predictive K48 finite geometric index, codon-amino-acid annotation layers, candidate-priority scoring, risk quarantine, no-call routing, blind candidate freeze rules, and external validation replay slots. Across the v1601-v2000 audit segment, 62,640 of 62,640 computational governance checks passed.This work is intended as a mathematical and algorithmic structural-screening scaffold for pre-experimental candidate prioritization. It does not make claims about clinical efficacy, prescribing, dosage guidance, human safety, human use, or drug approval.
