Abstract Bacterial metabolites, NDP-heptoses, are now emerging as versatile modulators of host signaling. ADP-glycero-β-d-manno-heptose, the best-characterized member, functions as a pathogen-associated molecular pattern sensed by alpha-protein kinase 1 (ALPK1). Recent studies reveal that context-dependent ALPK1 activation integrates microbial cues with host immune and tissue responses.
