Key Points

  • The aim is to investigate the tetracycline resistance capabilities of the P-7 incompatibility group plasmid pD4A-46 in host strains.
  • Analyzed plasmid pD4A-46 from Pseudomonas fluorescens for tetracycline resistance.
  • Examined expression levels in P. putida KT2442 (pD4A-46) under tetracycline exposure.
  • Compared expression with and without tetracycline to identify resistance determinants.
  • Identified putative genes associated with tetracycline resistance within the plasmid.
  • No known resistance genes were found in pD4A-46, indicating non-specific resistance mechanisms.
  • Expression levels varied significantly under different conditions, suggesting active resistance pathways.

Abstract

The conjugative plasmid pD4A-46, belonging to the P-7 incompatibility group, obtained from an environmental Pseudomonas fluorescens strain, confers tetracycline resistance to the host cells. However, no known tetracycline resistance genes were found in pD4A-46. Potential tetracycline resistance determinants within the pD4A-46 plasmid were investigated by analyzing the expression levels in P. putida KT2442 (pD4A-46) cells grown with and without tetracycline. The results of this analysis identified putative genes that function as tetracycline resistance determinants.
