Dysbiosis preventing property of rice bran extract from glutinous black rice and germinated brown rice

Introduction: Dysbiosis is an intestinal disorder caused from imbalance populations of microbes species between health beneficial macrobiotics and pathogenic macrobiotics. The chronic persistence of this could lead to various gastrointestinal pathology related to infections such as diarrhea, food poisoning, dysentery. Therefore, the purpose of this research was to investigate the feasibility in using of fermented rice bran extracts for prevention and treatment of dysbiosis. Method: The effects of fermented rice bran (FRB) obtained from black rice (H7F) and germinated brown rice (G13F) on growth of intestinal probiotic (Lactobacillus delbrueckii subsp. lactis; LAB) and pathogenic representative bacteria (Escherichia coli; E. coli) were determined using broth dilution technique using complete formula (MRS1) and half formula nutrient broth (MRS2) and flow cytometry. Result: The results showed prebiotic properties of both H7F and G13F as found of growth promoting effect to LAB that obviously demonstrated in MRS2. In contrast, inhibition of E. coli growth of both H7F and G13F was differently detected, in which, H7F showed stronger inhibitory effect with significant concentration at 3. 125 mg/mL. In contrast, G13F exhibited inhibition on E. coli growth with significance at concentration of 50 and 100 mg/mL. This growth inhibitory effects found in both H7F and G13F was related to producing of lactic acid, a natural acid, by LAB in fermentation process Discussion and Conclusion: The overall results indicated the potential of FRB extract in applications for dysbiosis prevention and treatment. This data has supported FRB to be further developed as health promoting product. คุณสมบัติป้องกันการเสียสมดุลของจำนวนประชากรเชื้อจุลชีพโดยสารสกัดจากรำข้าวก่ำและข้าวฮางงอก บทนำ: สภาวะขาดความสมดุลของแบคทีเรียชนิดมีประโยชน์และชนิดก่อโรคในระบบทางเดินอาหาร (dysbiosis) ในระยะยาวอาจส่งผลกระทบต่อสุขภาพที่รุนแรงและอันตรายแก่สุขภาพ เช่น ภาวะท้องเสียหรือท้องร่วง อาหารเป็นพิษ บิด เป็นต้น ที่เกิดจากการติดเชื้อจุลชีพก่อโรคหลากหลายชนิด งานวิจัยนี้จึงมีวัตถุประสงค์เพื่อศึกษาแนวโน้มในการประยุกต์ใช้ประโยชน์สารสกัดรำข้าวหมักสำหรับปรับภาวะไม่สมดุลของแบคทีเรียชนิดมีประโยชน์และชนิดก่อโรคในระบบทางเดินอาหาร (dysbiosis) วิธีการศึกษา: โดยศึกษาผลของสารรำข้าวหมักที่เตรียมจากรำข้าวก่ำ (H7F) และรำข้าวฮางงอก (G13F) ต่อการเจริญของเชื้อแบคทีเรียชนิดดี (Lactobacillus delbrueckii subsp. lactis: LAB) และเชื้อก่อโรคที่พบในลำไส้ (Escherichia coli; E. coli) ด้วยวิธี broth dilution technique ในอาหารเลี้ยงเชื้อแบบเต็มสูตร (MRS1) แลแบบครึ่งสูตร (MRS2) และวิธี flow cytometry ผลการศึกษา: พบว่า H7F และ G13F มีคุณสมบัติเป็นพรีไบโอติก ส่งเสริมการเจริญเติบโตของเชื้อ LAB โดยเห็นผลชัดเจนในอาหารเลี้ยงเชื้อสูตร MRS2 และสามารถยับยั้งการเจริญของเชื้อ E. coli ได้แตกต่างกัน โดย H7F แสดงการยับยั้งที่สูงกว่า ที่ความเข้มข้น ๓. ๑๒๕ มิลลิกรัมต่อมิลลิลิตร ซึ่งเป็นความเข้มข้นที่ส่งเสริมการเจริญของเชื้อ LAB ได้ด้วย ส่วนสารสกัด G13F พบการยับยั้งการเจริญของเชื้อ E. coli อย่างมีนัยสำคัญ ที่ความเข้มข้น ๕๐ และ ๑๐๐ มิลลิกรัมต่อมิลลิตร และการยับยั้งการเจรญิ ของเชือ้ E. coli ของสารสกดั รำขhkวหมกั จากขhk;ก่ำและข้าวฮางงอก น่าจะมีผลมาจากการที่เชื้อ LAB สร้างกรดธรรมชาติ เช่น กรดแลกติก ในระหว่างการหมักข้าว วิจารณ์ และสรุปผลการศึกษา: ผลการศึกษานี้แสดงให้เห็นศักยภาพของสารสกัดรำข้าวหมักในการปรับสมดุลปริมาณเชื้อจุลชีพในระบบทางเดินอาหาร ซึ่งข้อมูลนี้จะสนับสนุนพัฒนาสารสกัดรำข้าวหมักให้อยู่ในรูปผลิตภัณฑ์เสริมสุขภาพต่อไป
